Few dialogue examples:

Tanaka: Shitsureishimasu.
Sakura: Shitsureishimasu.
Miyagi: Doozo kochira e.
Tanaka, Sakura: Arigatoo gozaimasu.
Miyagi: Kuruma de kimashita ka.
Tanaka: iie, chikatetsu de kimashita.
Tanaka-san wa kin-yoobi ni Sakura-san to chikatetsu de Miyagi-san ni ikimashita.


Miyagi: Mr. Tanaka, Ms. Sakura, please come in.
Tanaka: Excuse me.

On Friday, Mr. Tanaka went with Ms. Sakura to Mr. Miyagi by subway.

Ohairikudasai: please come in
doozo kochira e: come right this way
de: by means of(particle indicating means)
chikatetsu: subway
kin-yoobi: friday
ni: at, on, in(particle indicating time)

The function of the particle de( by means of), which follows nouns, is to express means of the conveyance.
ex. basu de; by bus
takushii de: by taxi
kuruma de: by car
hikooki de: by plane
But to say by foot, use aruite, e.g., aruite kimashita, i walked here.
To ask the means by which someone will go somewhere, use nan de:
ex. nan de ikimasu ka. how will you go?
Basu de ikimasu. i'll go by bus.
Unlike relative time expressions; like ashita(tomorrow), raishuu(nextweek), kongetsu(this month) and kyonen(last year), which generally do not take particles, specific time expression take the particle ni.
ex. 5-ji ni: at 5:00
do-yoobi ni: on Saturday
12-nichi ni: on the twelfth.
2008-nen ni: in 2008
